چکیده
Dehydroabietic acid (DHA) is an analog of abietic (AA) for cardiovascular disease prevention, known to act pharmacologically against aging, inflammation, bacterial infections and cancer. The current research investigated the molecular mechanisms DHA on adhesiveness endothelial leukocytes activation NF-κB in TNF-α treated HAEC ApoE−/− mice experimental atherosclerosis. HAECs were tested toxicity using MTT assay at various concentrations. cell-surface expression CAM leukocyte adhesion molecule-1, ICAM -1, or human VCAM-1 was determined by ELISA test, followed western blot analysis. Endothelial cell-leukocyte involving U937 cells carried out determination p65 expression. fed with a high cholesterol diet every day oral administration (10 20 mg/kg/day). (5 10 mM) substantially reduced (p < 0.05) (U937) cell lines binding activated aortic cells. assays 32P-labelled NF-kB as probe demonstrated that pre-treatment decreased shifted bands density following stimulation TNF-α. Nuclear extracts immune assessment fluorescence staining showed significant decrease concentration nuclei Together these findings indicate inhibits nuclear trans localization TNF-α-induced p65, thus considerably suppresses expression, which contributes lower adherence suggesting helps preventing inflammatory atherosclerosis vivo.

Osteopontin deficiency attenuates atherosclerosis in female apolipoprotein E-deficient mice.
OBJECTIVE Osteopontin (OPN), a noncollagenous adhesive protein, is implicated in atherosclerosis, in which macrophages within atherosclerotic plaques express OPN. However, it is not known whether the elevated OPN expression is a cause or result of atherosclerosis. METHODS AND RESULTS We generated mice that lacked OPN and crossed them with apolipoprotein (apo) E-deficient mice and analyzed the...
متن کاملPPARgamma1-induced caveolin-1 enhances cholesterol efflux and attenuates atherosclerosis in apolipoprotein E-deficient mice.
OBJECTIVE Caveolin-1 (Cav-1) may positively or negatively influence the development of atherosclerosis, depending on the cell type and the metabolic pathways regulated by this protein. We investigate the effects of Cav-1 on cholesterol efflux in RAW264.7 infected with AdPPARgamma1 and whether Cav-1 could attenuate established atherosclerotic lesions in PPARgamma1-treated apoE-deficient mice. ...
متن کاملResveratrol protects against diet-induced atherosclerosis by reducing low-density lipoprotein cholesterol and inhibiting inflammation in apolipoprotein E-deficient mice
Objective(s):Resveratrol (RES) is a polyphenol compound that has been shown a promising cardioprotective effect. However, some reports have yielded conflicting findings. Herein, we investigated the anti-atherosclerotic effects of RES in apolipoprotein E (apo E)-deficient mice on a high cholesterol diet. Materials and Methods: Firstly, atherosclerosis was induced by feeding a high cholesterol di...
متن کاملAngiopoietin-2 attenuates angiotensin II-induced aortic aneurysm and atherosclerosis in apolipoprotein E-deficient mice
Angiogenesis and inflammation are implicated in aortic aneurysm and atherosclerosis and regulated by angiopoietin-2 (Angpt2). The effect of Angpt2 administration on experimental aortic aneurysm and atherosclerosis was examined. Six-month-old male apolipoprotein E deficient (ApoE-/-) mice were infused with angiotensin II (AngII) and administered subcutaneous human Fc-protein (control) or recombi...
متن کاملDBZ (Danshensu Bingpian Zhi), a Novel Natural Compound Derivative, Attenuates Atherosclerosis in Apolipoprotein E–Deficient Mice
BACKGROUND DBZ (Danshensu Bingpian Zhi), a synthetic derivative of a natural compound found in traditional Chinese medicine, has been reported to suppress lipopolysaccharide-induced macrophage activation and lipid accumulation in vitro. The aim of this study was to assess whether DBZ could attenuate atherosclerosis at early and advanced stages. METHODS AND RESULTS The effects of DBZ on the de...
